We are a clinical-stage biotechnology company developing a portfolio of internally invented, proprietary and wholly-owned oral inhibitors of NOD-like receptor protein 3, or NLRP3. NLRP3 is the upstream driver of sterile inflammation – inflammation that is caused by damage signals, such as cholesterol crystals or misfolded proteins, rather than by infection. NLRP3 activation directly stimulates the production of interleukin-1 beta, or IL-1ß, and interleukin-18, or IL-18, which are potent pro-inflammatory cytokines. When NLRP3 becomes chronically activated, it stimulates this cascade of inflammatory mediators that drive the development and progression of cardiovascular and neuroinflammatory disease. This phenomenon of chronically elevated inflammation that persists and contributes to disease progression even when other known risk factors are well controlled, is known as RIR. By directly inhibiting NLRP3 upstream, we aim to broadly reduce the activity of the entire sterile inflammation pathway, thereby reducing RIR and modifying the course of cardiovascular and neurologic diseases.
